Title: Spatial transcriptomics of human liver to characterize senescence with aging and fibrosis

Overview: Cellular senescence is implicated in the accumulation of fibrosis and progressive liver dysfunction with aging and disease. To characterize liver senescence in normal and diseased liver, we compared two spatial transcriptomics platforms, Nanostring GeoMx and 10X Genomics Visium. Serial tissue sections from young/healthy and old/diseased human livers were evaluated with each platform. A hypothesis driven approach was used with the GeoMx platform focused on the liver anatomical structures to compare senescence markers across tissues. A spatial discovery approach was used with the Visium platform to discover senescence spots with upregulation of senescence associated genes and increased senescence associated secretory phenotype (SASP) expression in the surrounding tissue. Both platforms identified increased senescence in old/diseased liver that spatially maps to fibrotic areas.
