Miltenyi Tyto Cell Sorter

Overview

The MACSQuant Tyto by Miltenyi is a next-generation, benchtop cell sorter equipped with 3 lasers, which allows for 10-parameter cell sorting. The sorting process occurs exclusively within the disposable, fully closed MACSQuant Tyto Cartridge, which keeps samples contamination-free and prevents aerosol and droplet formation. Unlike during conventional droplet sorting procedures, cells do not experience high pressure, they are not decompressed, and no charge is applied.

The UMGC offers the MACSQuant Tyto as a DIY instrument, giving researchers more options for flow-based collection, either upstream of UMGC analytical services or simply as a standalone flow cytometry service. Common applications are preparations of cell suspensions for single-cell or bulk RNA-seq workflows.

Training

At the required 4-hour training session, a UMGC scientist will guide a researcher through a "real" sort on the Tyto instrument, using viable cells provided by the researcher. The UMGC will train on instrument operation, maintenance, and clean-up protocols, and the researcher will be responsible for gating decisions, antibodies, etc. for the "real" sort training session.

Reserving the Tyto

  • Reservations are for a fixed 2-hour DIY session at $177.41/session.
  • Timeslot options are Monday - Friday either 9:00 am - 11:00 am or 11:00 am - 1:00 pm to enable two users per day.
  • The first scheduled reservation needs to be within 6 months of training.
  • Tyto location: 3-230E CCRB

Contact single-cell@umn.edu to reserve a 2-hour timeslot.

Purchasing a Cartridge

The UMGC purchases the Tyto cartridges in bulk and will provide individual cartridges to researchers at a cost. Researchers are not permitted to bring their own cartridges.
