Mission Bio Tapestri Single Cell


Overview

The single-cell multimodal workflow on the Tapestri platform by Mission Bio has the ability to quantitatively link genotypic and phenotypic data with future potential in biomarker identification and, ultimately, link to therapeutics. 

With DNA as a foundation, the proprietary two-step microfluidic workflow enables the simultaneous detection of analytes, including SNVs, CNVs, and proteins from the same cell, across thousands of single cells. Each reaction in the two-step workflow is discretely optimized: isolation of DNA from the cell and oligo-conjugate antibodies in the first step, followed by targeted multiplex PCR in the second step.

Whether a researcher is new to single-cell sequencing or one of our experienced users, we can help determine which single-cell technology will best meet your experimental goals. The UMGC requests that researchers contact us at single-cell@umn.edu for consultation on all incoming single-cell projects.

Pricing

Targeted cells/nuclei counts and the interrogating catalog or custom panels are project-specific and will determine the sequencing required and the associated cost. Project cost varies with the following factors:

  • Target panel (predesigned or custom)
  • Number of target cells (affecting the sequencing cost)
  • Service selected (self-service or full-service)


 The Tapestri instrument has two service options: 1) self-service (researcher-operated) in the UMGC CoLab, giving researchers convenient access to cutting-edge single-cell technologies that work around their project timeline, or 2) full-service (UMGC-operated) will be available Fall 2024.

Panels

Mission Bio provides a portfolio of pre-designed panels covering the most commonly implicated genes in hematologic malignancies and solid tumors. For more flexibility, researchers can create a custom panel based on current designs or from scratch for human, mouse, or other genomes. Panel options:

  • Hematologic Malignancy Protein Panels
  • Solid Tumor DNA Panels
  • Custom Panels for human, mouse, or any other reference genome


Oligo-conjugated antibodies from BioLegend can be combined with DNA panels to enable concurrent measurement of proteins.

Getting started


A planning meeting is needed to start a single-cell project to review your experimental goals, provide guidance on experimental design, and go over project logistics like turnaround estimates, scheduling, panels, and cell concentration requirements.
